蒙药特木仁-塔拉哈-5汤中铁对小鼠的毒性研究

摘要:

目的 研究蒙药特木仁-塔拉哈-5汤中铁的毒性。方法 测定特木仁-塔拉哈-5汤中的总铁和二价铁含量,并通过小鼠实验观察不同剂量特木仁-塔拉哈-5汤、特木仁-塔拉哈-5汤简化方、铁屑和铁粉连续灌胃14 d对小鼠的影响,观测指标包括一般状态、体质量、血常规、血清铁蛋白、尿液铁和锌含量、组织病理学等指标。结果 特木仁-塔拉哈-5汤和铁屑中总铁含量分别为16.3%和69.6%,二价铁含量分别为0.082%和0.323%。小鼠实验结果显示,各组小鼠精神状态良好,体质量无显著变化。高剂量特木仁-塔拉哈-5汤和铁屑组小鼠出现红细胞数量和血红蛋白含量下降,白细胞、淋巴细胞和中性粒细胞数目升高,以及血清铁蛋白含量上升,但未观察到明显的组织病理学改变和铁沉积。结论 特木仁-塔拉哈-5汤未导致机体铁代谢失衡,具有较好的安全性。

Abstract:

Objective To study the toxicity of iron in Mongolian medicine Temuren-Talaha-5 decoction. Methods The total iron and ferrous iron contents in Temuren-Talaha-5 decoction were determined. Mouse experiments were conducted to observe the effects of different doses of Temuren-Talaha-5 decoction, its simplified formulation, iron filings, and iron powder after continuous gavage for 14 days. Observation indicators included general condition, body weight, routine blood tests, serum ferritin levels, urine iron and zinc contents, and histopathological examination. Results The total iron contents in Temuren-Talaha-5 decoction and iron filings were 16.3% and 69.6%, respectively, while the ferrous iron contents were 0.082% and 0.323%, respectively. The mouse experiments showed that the mental state of the mice in all groups was good, with no significant changes in body weight. Mice in the high-dose Temuren-Talaha-5 decoction and iron filings groups exhibited a decrease in red blood cell count and hemoglobin content, an increase in white blood cell, lymphocyte, and neutrophil counts, as well as an increase in serum ferritin levels. However, no significant histopathological changes or iron deposition were observed. Conclusion Temuren-Talaha-5 decoction did not cause iron metabolism imbalance in the body and demonstrated good safety.
