目的 探讨miR-29b对人脐静脉内皮细胞(HUVECs)增殖与凋亡的影响。方法 实时定量PCR(q RT-PCR)法检测HUVECs中miR-29b的表达水平;HUVECs分别转染miR-29b mimics和miR-29b inhibitor后,MTT法检测HUVECs增殖活力、Hoechst 33342荧光染色法检测HUVECs的凋亡;生物信息学预测miR-29b的靶基因,双荧光素酶报告基因实验和Mcl-1蛋白表达检测实验验证其靶向关系;Western blot法、MTT法、Hoechst 33342荧光染色法分别检测Mcl-1质粒转染组和Mcl-1质粒与miR-29b mimics共转染组的Mcl-1蛋白表达、HUVECs增殖与凋亡情况。结果 miR-29b表达于HUVECs中;与对照组相比,转染miR-29b mimics的HUVECs增殖活力降低,细胞凋亡数增加,而转染miR-29b inhibitor的HUVECs增殖活力增加,细胞凋亡数减少;Mcl-1被确定是miR-29b的直接靶标;Mcl-1的过表达减弱了miR-29b对HUVECs增殖和凋亡的调控作用。结论 miR-29b通过靶向Mcl-1抑制HUVECs的增殖并促进其凋亡。
Abstract:Objective To investigate the impact of miR-29b on proliferation and apoptosis of human umbilical vein endothelial cells(HUVECs). Methods The expression level of miR-29b in HUVECs were detected by q RT-PCR.miR-29b mimics and inhibitor were transfected into HUVECs,respectively.The proliferation of HUVECs were analyzed using MTT assay, Heochst 33342 staining was used to detect the apoptosis of HUVECs.The target gene of miR-29b was predicted by bioinformatics analysis, the dual-luciferase gene reporter assay and the protein expression of Mcl-1 by Western blotting were applied to verify the targe ring relationship.Western blotting,MTT assay,Heochst 33342 staining were used to detect the protein expression level of Mcl-1,the proliferation and apoptosis in cells transfected with Mcl-1 plasmid or the Co-transfection of Mcl-1 plasmid and miR-29b mimics,respectively. Results miR-29b was expressed in HUVECs.Compared with the Control Group,The proliferation of HUVECs in miR-29b mimics group were decreased,while The amount of Apoptotic HUVECs were increased. The proliferation of HUVECs in miR-29b inhibitor group were increased,whereas The amount of Apoptotic HUVECs were decreased. Mcl-1 was identified as a target of miR-29b that could directly bind to it. Overexpression of Mcl-1 attenuated the regulatory effects of miR-29b on proliferation and apoptosis in HUVECs. Conclusion miR-29b could inhibit the proliferation and promote the apoptosis of HUVECs by targeting Mcl-1.
